Construction of a Shuttle Vector for Heterologous Gene Expression in Escherichia coli and Microalgae Anabaena
نویسندگان
چکیده
The construction of an integrative shuttle expression vector and potential utility was reported in Escherichia coli and Anabaena (Nostoc) sp. strain PCC 7120. The vector comprised of the following elements: (a) an intergenic non-coding region from Anabaena to facilitate its genomic integration (b) a strong functional PpsbAI promoter from Anabaena for desired gene expression and (c) neomycin phosphotransferase gene with its own promoter for the selection of transformants. The constructed vector pAnFP was evaluated by cloning, transfer and expression of the gfp gene encoding green fluorescent protein. When the E. coli and Anabaena sp. strain PCC 7120 were transformed, intensive green fluorescence produced by the products of GFP protein was observed. This result indicated that the integrative shuttle vector pAnFP can be promisingly used in genome transformation for expression of heterologous genes in E. coli and microalgae such as Anabaena and Nostoc strains.

Rumen bacterial strains can potentially be manipulated to perform functions different from wild type species. The most numerous species of bacteria in the rumen and gut are species of the familyBacteroidetes, whichcan have the potential for genetic modification for enzyme production. One of the genetic manipulation of rumen bacteria can perform for production of starch digestive enzyme for the ...

Infectious Pancreatic Necrosis Virus (IPNV) is a member of the family Birnaviridae that has been linked to high mortalities in salmonids. Bacterial based systems as live vectors for the delivery of heterologous antigens offer a number of advantages as vaccination strategies. VP2 is a structural viral protein of IPNV with immunogenicity effects. In this study IPNV was isolated from diseased fry ...

Background: Corynebacterium glutamicum (C. glutamicum) is a potential host for the secretory production of the heterologous proteins. However, to this date few secretion-type gene expression systems in C. glutamicum have been developed, which limit applications of C. glutamicum in a secretory production of the heterologous proteins. Objectives: In this stu...

متن کاملSynthesis and Expression of Modified bFGF Gene in Escherichia coli Cells
A new strategy for construction of synthetic gene encoding human basic fibroblast growth factor comprising DNA annealing-ligation and augmentation by polymerase chain reaction was introduced. The sequence of the gene and corresponding amino acid chain were modified in order to increase stability of the protein. First, 300 bp and 160 bp fragments of the gene were assembled from 18 oligonucleotid...
